Contamination Control Strategy

Annex 1 CCS Methodology

Our structured, risk-based approach to Contamination Control Strategy development — fully aligned with EU GMP Annex 1 (2023 revision) and TGA requirements.

What is a Contamination Control Strategy?

A Contamination Control Strategy (CCS) is a planned set of controls — derived from current product and process understanding — that assures the protection of the product from contamination. It is now a mandatory requirement under EU GMP Annex 1 (2023).

The CCS must be a holistic, living document that covers all aspects of contamination prevention — from facility design through to personnel behaviour and environmental monitoring. It is not a single document but a framework that links all your contamination controls together.

Our team has developed and implemented CCS programmes at major sterile manufacturing sites including CSL Behring (Melbourne), Novartis TechOps (Slovenia), and Sandoz. We bring practical, inspection-ready expertise to every engagement.

CCS Must Cover

Facility design and cleanroom classification
Air handling and HVAC systems
Equipment design and qualification
Utilities (WFI, clean steam, compressed gases)
Personnel gowning and aseptic technique
Environmental monitoring programme
Cleaning and disinfection procedures
Material and component controls
Process design and aseptic simulation
Pest control programme
Change control and deviation management
Supplier qualification

Our 5-Phase CCS Approach

A structured, risk-based methodology that delivers inspection-ready CCS documentation and implementation.

01

Gap Analysis

Systematic assessment of your current contamination control measures against EU GMP Annex 1 (2023) requirements. We identify gaps, prioritise risks, and define the remediation roadmap.

Deliverables
  • Annex 1 gap assessment report
  • Risk-ranked findings register
  • Remediation action plan with timelines
02

Risk Assessment

Formal contamination risk assessment using ICH Q9 principles — identifying contamination sources, routes, and controls across your facility, processes, and personnel.

Deliverables
  • Contamination risk register
  • Risk heat maps by zone and process
  • Control effectiveness ratings
03

CCS Document Development

Authoring the formal Contamination Control Strategy document — a living document that defines your holistic approach to contamination prevention across all critical zones.

Deliverables
  • Contamination Control Strategy (CCS) document
  • Control measure matrix
  • Environmental monitoring programme
04

Implementation Support

Hands-on support to implement the CCS across your site — updating SOPs, training personnel, qualifying equipment, and establishing monitoring programmes.

Deliverables
  • Updated SOPs and work instructions
  • Personnel training records
  • Equipment qualification support
05

Ongoing Review & Maintenance

Annual CCS review and update to reflect process changes, regulatory updates, and environmental monitoring trend data.

Deliverables
  • Annual CCS review report
  • Trend analysis and CAPA
  • Regulatory change monitoring

EU GMP Annex 1 (2023) — Key CCS Requirements

The revised Annex 1 (effective August 2023) explicitly requires manufacturers to develop and maintain a formal Contamination Control Strategy. Section 4.1 states: "The manufacturer should have a contamination control strategy (CCS) implemented across the facility to define all critical control points and assess the effectiveness of all the controls (design, procedural, technical and organisational) and monitoring measures employed to manage risks associated with contamination."
